I want to use the "Retina Mode" setting in Screen Options (Mac Driver) because without it, the game I am playing looks blurry.
The "Retina Mode" setting makes the game/pixels look crisp, but it shrinks the entire game in half - making it too small to play.
From here, is there a way I can upscale the window while also retaining the sharpness of the Retina Mode feature?

You can change the font scaling in winecfg but really enabling retina mode just disabled macOS building scaling.
I remember seeing a comment from a CodeWeavers dev saying don’t bother with retina mode.

That's a bummer that Retina Mode has that effect.
