I am very new to this kind of thing. I got Manga Studio 5 running almost perfectly under Wine 1.6.2, except for one problem: whenever I press any modifier key like Ctrl or Shift, the cursor starts flickering between the regular Wine cursor and the drawing cursor the program is supposed to have, and I am unable to interact with the canvas. The problem goes away when I alt+tab out of Manga Studio and back again, but it returns whenever I press Ctrl or Shift.
I would try changing the tablet settings from WinTab to Tablet PC, but the latter is greyed out for me. If there are any drivers I could install to get this option I would be willing to give it a shot.

However, that report is old, and so is your Wine version. Retest in the current development release, and leave a comment in the bug report stating whether the problem is fixed or still present.

I've upgraded to 1.7.50 and now the program crashes and gives the "experienced a serious problem and needs to close" error instantly upon startup.

If you didn't reinstall to a clean wineprefix after you upgraded, try that. If that's not it, run a regression test and file a bug for the crash. http://wiki.winehq.org/RegressionTesting
